The Tykes arrive on the back of a morale-boosting 3-1 victory over Bradford City at Oakwell last weekend and boast a decent away record this season.
Posh go into the clash following a 4-1 away win at Crawley Town, a display that won the Performance of the Week.
Defender Kgosi Ntlhe and striker Kyle Vassell are ruled out through injury, but Nathaniel Mendez-Laing is expected to return to the squad after playing an hour in the development side's victory over Leyton Orient in midweek.
Boss Darren Ferguson commented: 'I watched Barnsley last week and they have a good squad. Their back line is very experienced at this level and ahead of that they have good footballers and a lot of pace in attack.
'They came down from the Championship last term and it does take a little while to adjust to the level. They are certainly doing that now and they will come here full of confidence.
'It has been a good week for us, the lads are in good spirits and we are looking forward to the game,' Ferguson said.
Barnsley boss Danny Wilson has said they would be interested in bringing Brighton striker Chris O'Grady back to the club.
The 28-year-old left Oakwell for the Seagulls in the summer but has so far failed to score in seven appearances.
'We didn't want Chris to go in the first place so if he was available then we would ask the question. But as far as I know he is still a Brighton player and will remain there.'
He added: "We wouldn't be the only club asking about him if he were available.'
