Darren Ferguson's men have won every game they have played this season including an emphatic 5-1 win at League One side Colchester United in the first round of the competition.
The Royals were beaten 1-0 at the weekend, but Nigel Adkin's men will believe they have the squad to cope with the demands of the Championship and the Cup competitions.
Ferguson hinted after the win over Tranmere Rovers that he is set to make changes to his starting line-up with Grant McCann and Kgosi Ntlhe under threat.
'I will definitely look to make changes against Reading. We have a squad and I have to utilise it. Jack Payne will definitely be involved so I may look to give Grant a rest in midfield.
'Mark Little was excellent when he came on so I could switch Craig Alcock to left-back and play him at right back. I have to be careful with Kgosi because he is a young lad and has been out for a long while in the past, so I need to take that into consideration,' Ferguson said.
Posh and Reading have met regularly in the competition with the Royals winning 3-2 at the Madesjki Stadium last season. Paul Taylor and Lee Tomlin were on target for Posh.

The adult ticket price will rise to £20 for adults on the day, but the concession and junior price will remain. The Executive Suite tickets are available for £25 for adults and £15 for concessions.
Season ticket holder seats will be held until 5pm on the day of the game. Access can be added to those with Smartcards.
